Les effets secondaires indesirables des medicaments Vioxx (Merck) et Bextra (Pfizer) ou Celebrex (Pfizer) eventuellement pourraient conduire une collaboration etroite et rapide entre Nicox, Pfizer et Merck pour accelerer les developpements des molecules debutants de phase 3 ???

Convergence des nouvelles (Nicox - Pfizer - Merck) avec le graphe de Nicox, demain si on arrive a casser le triple top = 3,95 on pourra atteindre 4,1x
une collaboration plus etroite avec Pfizer et Merck ou qui sait une eventuelle opa en preparation ???? LOL  une poule aux oeufs d'or ?  attention je me trompe souvent

citation : hello rahan, les regles de weinstein sont formidables. meditons ceci : la communication a bcp evolue depuis 1994 grace a l'internet, un courrier prend un jour a un mois pour arriver a son destinataire avant les emails, les ordres de bourse sont bcp moins fluides que maintenant, tout le monde peuvent passer des ordres par son courtier en ligne. Donc suivant moi il faut appliquer la notion " Espace - Temps" aux phases de Weinstein. C'est à dire les durees des phases à l'age de l'internet sont beaucoup plus courtes qu'avant. la patience et le longueur du temps diminuent aussi, lol bon trade

pour une vision et une investigation sous le dernier decan de la "Balance" NicOx's NO-ASA, NCX 4040, reduces the developement of intestinal tumors Cancers of the colon and rectum remain responsible for 56,000 deaths every year in the US, second only to lung cancer. Surgery remains the main form of treatment and is often the only treatment used in early-stage disease. Current surgical practices can however cure only 40%-50% of early-stage cases thereby driving the development of pharmaceutical approaches to colorectal cancer. The global colorectal cancer market was estimated to be worth approximately US$1 billion in 2001, of which 60% of the revenues were derived from the US. The market has been predicted to rise further to US$2.5-3 billion within a five-year period. Camptosar currently dominates the US colorectal cancer market, but severe diarrhea as a side effect is a clear drawback for this drug (for a full analysis of therapeutic and market opportunities for colon cancer click here). Early detection or prevention of the colorectal cancer is particularly important and the observation that non-steroidal anti-inflammatory drugs (NSAIDs) prevent the development of colon and other cancers in humans has revolutionized the approach to colon cancer prevention. Aspirin (ASA) has emerged as the prototypical chemoprevention agent against colon cancer. Current efforts are now being directed towards improving efficacy and reducing side effects. Of note, the NSAID class produces gastro-intestinal side effects precluding their use in at risk individuals. Nitric oxide (NO) facilitates the repair of injury to the gastrointestinal tract by stimulating mucus secretion and by regulating the blood flow in the capillaries of the gastrointestinal tract and the mucus membrane. The multiple indications of NO therapeutics including gastroprotective activity has led to a surge in pharmaceutical activity (click here for an evaluation of NO therapeutics). Grafting NO to NSAIDS combines the chemopreventive properties of traditional NSAIDs against cancer with enhanced safety. Furthermore, the chemopreventative activity of NSAIDs appears to be increased by the presence of the NO. NO-ASA is particularly potent as a chemopreventive agent and the gastrointestinal toxicity of NO-ASA is minimal in humans. The French company NicOx is one of the leaders in developing NO derivatives. One of their candidates, NCX 4040 is currently in preclinical development for the prevention of colon cancer. In their recent BBRC paper, Dr Basil Rigas' group from the American Health Foundation Cancer Center in collaboration with Dr Khosrow Kashfi at City University of New York Medical School and others report on the ability of this novel nitric oxide-donating derivative of aspirin to inhibit the development of gastrointestinal tumors in the Min mouse. These mice spontaneously develop small intestinal tumors and are commonly used in studies designed to assess molecules designed to prevent colon carcinogenesis. Consistent with human toxicity data, at the doses of NCX 4040 employed, Williams et al failed to observe evidence of GI toxicity however the number of tumors identified was reduced by 59%. These results demonstrate the tumor inhibitory properties of NCX 4040. NCX 4016, another nitric oxide-donating derivative of ASA in development by NicOx, has completed Phase IIa evaluation in patients with symptomatic peripheral arterial obstructive disease (PAOD). In a phase I study NCX 4016 displayed an optimal gastric safety profile compared with aspirin and according to the NicOx website a US $3.3 million grant has been awarded by the National Cancer Institute of the NIH to the University of Michigan to conduct a clinical trial of nitric oxide-donating aspirin as a chemopreventive agent. The results of this trial along with further data on NCX 4040 are eagerly awaited. Mundell HealthDay Reporter TUESDAY, Oct. 19 (HealthDayNews) -- An aspirin that is "thousands of times more powerful" than traditional forms of the drug but has no gastrointestinal side effects looks promising in animal studies, researchers say. The drug, called nitric oxide-donating aspirin, or nitroaspirin, appears to help prevent colon cancer in mice without raising the incidence of gastrointestinal bleeding, researchers reported Oct. 19 at the American Association for Cancer Research meeting in Seattle. Although the findings are "preliminary," the drug may be also help protect against cardiovascular disease and ease arthritis pain, said researcher Dr. Basil Rigas, chief of the Division of Cancer Prevention at SUNY Stony Brook, New York. "My laboratory has been working with this compound for the last five years and it has two striking features -- one, it's much more potent than regular aspirin, and secondly, it's much safer," he said. Clinical trials involving the use of nitroaspirin in 240 patients at high risk for colon polyps and colon cancer are expected to begin "over the next few months," he added. Doctors have long known that aspirin -- a drug first discovered more than 100 hundred years ago -- prevents or treats a number of common ailments. However, aspirin, like other members of the non-steroidal anti-inflammatory (NSAID) family of drugs, tends to raise risks for gastrointestinal bleeding in long-term users. In fact, popular cox-2 arthritis medications such as Celebrex, Bextra and Vioxx -- the last of which was recently withdrawn from the market due to increased risk of heart attack or stroke -- were successful because they relieved pain without the gastrointestinal risk inherent in other NSAIDs. Many NSAIDs, including aspirin, also seem to help lower risks for colon cancer. "That's actually where some of the initial data on colon cancer came from, " said Dr. Durado Brooks, director of prostate and colorectal cancer at the American Cancer Society. "People who were taking these drugs for arthritis were coincidentally noticed to have a lower likelihood to have [colon] problems." However, with regular aspirin's gastrointestinal side effects, scientists have been busy looking for alternatives. Enter nitroaspirin. "It's similar to traditional aspirin, but different in one way, in that the molecule of aspirin has been modified to release nitric oxide (NO)," Rigas explained. "Nitric oxide is a very important molecule that has multiple effects within the cardiovascular system, the respiratory system and, we are finding, against cancer." In their study, Rigas and his colleagues found that mice engineered to have a high risk of colon tumors that were given nitroaspirin daily for three weeks displayed a 59 percent reduction in tumors compared with untreated mice. And there were no signs of increased gastrointestinal toxicity compared to placebo. The study was funded by a grant from the National Cancer Institute. Brooks said the findings "look very promising, and it's an exciting concept." But he stressed that, "we're a long way from being able to say anything about nitroaspirin's true value, given that what we've looked at is an animal study." He also noted that drugs that look safe in the short-term can lose their appeal when long-term data arrives. "Remember, cox-2 inhibitors [such as Vioxx] have been used for a long time and were felt to be safe and effective -- they were a form of 'safer aspirin,' too," he said. "So who knows? Maybe somewhere down the line we may find out about the side effects of nitroaspirins. We just don't know right now." Still, Brooks remains cautiously optimistic. The drug could well prove to be "a single agent that helps decrease the incidence of heart disease as well as decreasing colon cancer risk," he said. "It might even be a chemotherapeutic agent for someone who has a colon polyp identified -- you give them this to make it go away." Rigas agreed the findings are preliminary, but he said the drug's potential impact could be enormous. "Our investigators have studied this for prophylaxis against heart disease, also against arthritis," he said. "It's up to 5,000 times more powerful than regular aspirin, and a lot safer."
